Glycan shielding on the Envelope protein of Zika and Dengue virus against broadly neutralizing Envelope Dimer Epitope (EDE) specific antibodies revealed by hybrid mass spectrometry
Recently, potent cross-neutralizing antibodies were characterized that recognize the so-called envelope dimer epitope, EDE, on the Dengue closely related Zika envelope glycoprotein. The structure of EDE-antibody in complex with the envelope antigen shows the presence of N-glycans at the antibody-binding site. We demonstrate that EDE-antibodies preferentially bind DENV-2 and ZIKV E subunits, decorated with truncated chitobiose structures at N67 and N154 sites respectively, indicative of strong N-glycan shielding. Our findings can play a positive role in future subunit vaccine design.
