Analysis of CFTR folding using domain-specific antibodies
The folding pathway of polytopic membrane proteins is relatively unknown, despite their critical role in a variety of cellular functions and linkage to disease. We studied the folding of the ABC transporter CFTR, which is linked to cystic fibrosis (CF). Using biochemical methods, we investigated how domains fold, and how and when domain-domain interactions are established. Our results enhance current understanding on CFTR folding and may establish a paradigm for the folding pathway of structurally related ABC transporters.
